Benzylmercaptotetrazole (BMT, BTT) is a novel activator for use in chemical synthesis of either DNA or RNA. BMT has been shown to outperform ETT, DCI and Tetrazole with respect to coupling times, coupling efficiency, lower consumption of phosphoramidites and a lower appearance of n - 1 impurities for both DNA and RNA. For RNA, this allows the routine synthesis of longer oligonucleotides over 40 base pairs in length.
Being non-explosive, BMT is significantly safer than 1H-tetrazole. Furthermore, the coupling efficiencies using BMT are largely independent of the 2'-O-protected phosphoramidite monomer used in RNA synthesis and can be used with TBDMS, O-Methyl, TOMŽ or ACEŽ.

Hyacinth BMT Solution (Benzylmercapto-1H-tetrazole in anhydrous acetonitrile) is a chemical activator for both DNA and RNA synthesis and is used as a substitute for 1H-Tetrazole, DCI and ETT. Hyacinth is available in 0.2 M, 0.25 M or 0.3 M concentrations.
For RNA synthesis it is compatible with both TBDMS, ACE and TOM protected monomers. It improves RNA coupling yields to 99%, requires only 3 minute coupling times, requires 50% less phosphoramidite for each coupling and is easy to handle. |
