ZetaCell Ni-NTA is a large cross-linked agarose bead, derivatized with nitrilotriacetic acid (NTA) ligand. When charged with nickel ions, the solid phase can be used to purify recombinant proteins containing polyhistidine (polyhistidine-tag) residues via their selective affinity to nickel. After washing, bound proteins are eluted under native or denaturing conditions with either a low pH buffer or with buffer containing imidazole or histidine. It is specifically designed for SMART™ IMAC chromatography.
SMART Chromatography™ uses proprietary ZetaCell solid phases. These are based on large-particle highly cross-linked agarose, which is surface-modified to provide the desired binding functionality. The beads have been designed to allow cells and cell debris from the bioreactor to pass unhindered through the packed column bed.
