SMART Chromatography™ NTA is packed with large cross-linked agarose beads, derivatized with metal-free nitrilotriacetic acid (NTA) ligand. When charged with bivalent metal ions, such as nickel, cobalt, copper, or zinc, the solid phase can be used to purify recombinant proteins containing polyhistidine (6xHis) residues via their selective affinity to chelated metal. After washing, bound proteins are eluted under native or denaturing conditions with either a low pH buffer or with buffer containing imidazole or histidine. It is specifically designed for SMART Chromatography™ IMAC protein purification.
SMART Chromatography™ uses proprietary ZetaCell solid phases. These are based on large-particle highly cross-linked agarose, which is surface-modified to provide the desired binding functionality. The beads have been designed to allow cells and cell debris from the bioreactor to pass unhindered through the packed column bed.
